I don't know for certain, but CHESS 4.5 won the Minnesota Open Championship in 1977, which probably required at least one win against a master. It then played in the closed championship and won one game, in a field that may well have been mostly masters. In 1978 IM David Levy lost game 4 of his match with CHESS 4.7 (a match he won 3.5 to 1.5), so 1978 at the latest was the first loss for a human master in a game played under tournament conditions.
